Lay everything flat on a clean surface. Wipe the vial's grey stopper with an alcohol swab.

Gentle mixing
Draw the bacteriostatic water, inject slowly along the glass wall — without aiming directly at the powder.

Dissolution
Do not shake. Gently roll the vial between your hands until the liquid becomes clear.

Understanding the calculation

The calculation relies on three simple steps, in order :

  1. Concentration = amount of peptide (mg) ÷ volume of water added (ml). This is the "strength" of your solution once reconstituted.
  2. Volume to draw = desired dose (mg) ÷ concentration (mg/ml).
  3. Syringe reading = volume (ml) × 100, since a U-100 insulin syringe is graduated at 100 units per milliliter, regardless of its physical size.

In practice: a 10 mg vial reconstituted with 2 ml of water gives a concentration of 5 mg/ml. To draw 2 mg, you need 0.4 ml — i.e. 40 units on the syringe.

Which syringe size should you choose ?

The rule is simple: always take the smallest syringe that can hold your volume. A 0.3 ml syringe (30 units) has its graduations more spaced out than a 1 ml syringe for the same real volume — making it easier to read precisely for small volumes. A 1 ml syringe becomes necessary as soon as the calculated volume exceeds 0.5 ml.

Why does the water volume change the result ?

The more bacteriostatic water you add, the more diluted the solution — so the larger the volume needed for the same dose. Conversely, less water gives a more concentrated solution and a smaller volume to draw. The choice of water volume is a matter of comfort reading the syringe, not a matter of dose.
